Biophysics
Bio-Rad NGC Fast Protein Liquid Chromatography Units
We utilize two Bio-Rad NGC units.
First is a multistep protein purification using
- affinity chromatography
- size exclusion chromatography (SEC)
- ion exchange desalting columns
includes 10 ml/min pumps, 4-wavelength (UV) and conductivity detection, automated sample injection, and sample pump.
Second is a part of a SEC-MALS purification and analysis system for molecular weight and multimeric determination. It consists of several devices coupled in line:
- FPLC (NGC Quest™ Plus 10, Bio-Rad)
- Dynamic Light Scattering Detector (DynaPro NanoStar, Wyatt)
- 3-angle Static Light Scattering Detector (miniDAWN™ TREOS®, Wyatt)
- RI Detector (RefractoMax 521, Analytisch) – refractive index detector
Dynamic Light Scattering Detector
Dynamic Light Scattering Detector (DynaPro NanoStar, Wyatt) is used for measurements of size, size distributions, and weight-average molar mass, as well as solution properties relevant to sample stability and the temperature of protein melting or aggregation.
While it is a part of a pipeline with Bio-Rad NGC Quest, it can be also used as a stand alone device using cuvette mode for fast sample measurements.
Microscale thermophoresis system
Microscale thermophoresis system (MST, Monolith® NT.115 Blue/Red, NanoTemper) for ligand and protein binding studies.
Plate reader
Plate reader (Infinite M200 PRO, Tecan) with absorbance, fluorescence and luminescence modes with an automated pipetting module for enzymatic assays and readouts.
Real-Time PCR System
Real-Time PCR (LightCycler® 96, Roche) instrument destined for thermal shift assays setup using.
Nanospectrophotometer UV-Vis
Nanospectrophotometer UV-Vis (DeNovix DS-11) is used for 0.5 – 1.0 microliters volume measurments of protein and nucleic acid samples.
Imaging System
Imaging system (ChemiDoc MP, Biorad) for analyzing gels and western blots.
X-Ray Crystallography
Automatic imaging and storage system
Automatic imaging and storage system (Rock Imager, Formulatrix) for up to 1000 crystallization plates, with UV, visual light and polarized light visualization modes.
High-quality stereo microscopes
High-quality stereo microscopes (SteREO Discovery V8 Zeiss, X2000 OptaTech, SZX16 Olympus) for visualization and manipulation of crystals.
High-throughput screening robot
Protein crystallization robot (Mosquito LCP, TTP Labtech) pipetting in nanoliters volume using single used tips, allowing for high-throughput automated crystallization approaches (2 or 3 sitting drop or hanging drop, Fig 1), including LCP (Fig. 2) for membrane proteins.
Conditions optimization robot
Conditions optimization robot (Dragonfly, TTP Labtech) for automated setup of crystallization condition screening.
X-ray diffractometers with LT attachments for cryo-measurements and setup for high-pressure studies
